Support data_ FOXD1 Promotes Dedifferentiation and Targeted Therapy Resistance in Melanoma by Regulating the Expression of CTGF.zip

In this manuscript, we show that FOXD1 is highly expressed in melanoma cells and is associated with a poor survival in patients with metastatic melanoma. FOXD1 can promote melanoma cells resistance to vemurafenib (BRAF inhibitor) or vemurafenib and cobimetinib (MEK inhibitor) combination treatment. whereas, loss of FOXD1 increases cell sensitivity to the targeted therapy. Interestingly, we identified the connective tissue growth factor (CTGF) to be a downstream factor of FOXD1. FOXD1 can promote cell resistance by regulating CTGF expression. Based on these findings, we conclude that FOXD1 might be a promising new diagnostic marker and a therapeutic target for the treatment of targeted therapy resistant melanoma.
