Macrophage, BMDM, pro-inflammatory, innate immunity
Label free quantification of proteins identified by LC-MS/MS based proteomics in untreated BMDMs and BMDMs exposed to pro-inflammatory conditions.
BMDMs were treated for 24 h with [20 ng/mL] interferon gamma (IFN-γ), [20 ng/mL] lipopolysaccharide (LPS), [100 ng/mL] Pam2CSK4, [100 ng/mL] Pam3CSK4, [1 µM] unmethylated cytosine-phosphate-guanosine-oligodeoxynucleotides (CpG-ODNs), [1 µg/mL] single-stranded RNA (ssRNA) polyuridylic acid (poly (U)), [1 µg/mL] double-stranded RNA (dsRNA) polyadenylic:polyuridylic acid (poly(A:U)) and [1 µg/mL] dsRNA polyinosinic:polycytidylic acid (poly(I:C)). BMDMs were lysed, trypsin-digested and resulting peptides analysed by LC-MS/MS with an orbitrap mass spectrometer.
LFQ values were generated after searching the ion peak intensities against the mouse proteome using MaxQuant software. Statistical analysis and data collation were performed in the R statistical programming environment.
