Screening of a Monoclonal Antibody Recognizing a Novel Conserved Epitope of ApxIVA and Its Application in a Blocking ELISA

Abstract:To establish a highly specific serological detection method for the ApxIVA protein, the N-terminal recombinant protein of ApxIVA was expressed in prokaryotic cells, and a murine monoclonal antibody, 1D5, was generated using hybridoma technology. It was identified that this monoclonal antibody recognizes a novel conserved epitope of ApxIVA. Subsequently, a soluble truncated protein containing the epitope peptide was successfully expressed. Based on this, a blocking ELISA method using the soluble truncated protein of APP ApxIVA was established. The results showed that this method has high sensitivity and specificity, providing an effective tool for the diagnosis of ApxIV-related diseases and antibody detection.
